This line is from the song "Supermodel" by RuPaul, from the album Supermodel of the World (1992).

Arguably Earth's top drag queen, RuPaul exploded onto the scene when this song hit the clubs and its video hit MTV. Basically, it tells the story of young "girl" who wants to be a supermodel and has to work it on the runway to make it happen. These days, RuPaul spends his time teaching the drag queens of the future on the competitive reality show Drag Race.

Where you've heard it

It's impossible that you haven't heard this line before, since RuPaul helped to turn "you better work" into a genuine idiom. It's even in Urban Dictionary, where it's defined as "an imperative phrase used to give praise or approval to another person." You might've also seen it shortened into "werk" or even "shwerk."
