Context

This line is from the song "Like a Virgin" by Madonna, from the album Like a Virgin (1984).

Madonna is not a virgin. She released a book of erotic photography in 1992 called Sex…in case you had any doubts about that.

So when she sings that she feels "like a virgin, touched for the very first time," she means that she's having feelings for a man that she's never felt before—and it's a four-letter word different from the ones she celebrates in Sex: L-O-V-E.

Where you've heard it

You've heard this song on Glee in an episode about teens losing their virginity, which means the creators didn't listen to the song at all.

It's also covered in Moulin Rouge, and Baz Luhrmann, who uses the song to refer to the courtesan Satine's newfound feelings of love and romance, clearly did listen to it.
